Identification and Expression Analysis of the PHR Gene Family in Pennisetum Purpureum

Abstract:The phosphate starvation response (PHR) factor plays a pivotal role in regulating phosphorus homeo stasis in plants,yet the PHRs in Pennisetum purpureum (elephant grass)remain unreported. This study analyzed the efects of low phosphorus (P)stress on the growth of elephant grass,identified the members of PHR family in elephant grass,and examined their transcription expression levels.Furthermore,the two key genes P/PHR/13 were cloned,and their expression patterns and subcellular localization were analyzed. The results demonstrated that low P stress significantly inhibited the growth of elephant grassA total of 42 PpPHRs were identified from the elephant grassgenome,with colliear gene pairs originating from whole-genome duplication or segmental duplication events. Transcriptome showed that 1 5PρPHRs were significantly upregulated by low P stress in elephant grass leaves(and/or roots),with P/PHR/13 exhibiting relatively high expression levels. Additionally, tissue-specific expression analysis revealed that P/PHR/13 had the highest expression levels in roots and stems,respectively. Quantitative PCR confirmed that low P treatment enhanced the expression of

P/PHR/1/3 in both roots and leaves. Subcellular localization indicated that both P/PPHRδ andPpPHR13pro teins were localized in the nucleus. This study provides candidate gene resources for elucidating the mechanisms underlying elephant grass's response to phosphorus deficiency.
