Abstract: With the gelatin of cod (Gadus morhua)skin prepared by water extraction method as the raw material,single factor test and response surface method are used to optimize the double-enzyme stepwise enzymatic hydrolysis process conditions of cod skin ACE inhibitory peptides. Combined with membrane separation method,the preparation process of cod skin ACE inhibitory peptides is established. The first step of enzymatic hydrolysis: enzyme type is hydrolase,enzyme addition amount is 4% , solid-liquid ratio is 1:20 , pH is 9,enzymatic hydrolysis temperature is 50∘C ,and enzymatic hydrolysis time is 4.5h ;the second step of enzymatic hydrolysis: enzyme type is flavourzyme,enzyme addition amount is 3% ,solid-liquid ratio is 1:20 , ΔpH is 6,enzymatic hydrolysis temperature is 40∘C ,and enzymatic hydrolysis time is membrane separation method:1o,3,1 kDa ultrafiltration membranes are used respectively for the separation of obtained enzymatic hydrolysates to obtain the target components. The IC50 value of ACE inhibitory rate of the product prepared under double-enzyme stepwise enzymatic hydrolysis process conditions is 1.46mg/mL , the total content of free amino acids is 71.04mg/100g ,and the polypeptides with the molecular weight below 1 ooo Da account for 99.5% ; the ACE inhibitory rate of collagen peptides from cod skin produced after 1 kDa membrane separation increases by 4.25 times. In this study,the preparation method of cod skin collagen peptides with high ACE inhibitory activity is established,and the prepared product can layamaterial foundation for the development of functional antihypertensive condiments and foods.
