Advances in methionine restriction for cancer therapy

Abstract: Methionine (Met),an essential amino acid,serves not only as a fundamental building block for protein biosynthesis but also as a critical node regulating cellular metabolic networks. Recent studies have revealed that various malignancies exhibit a distinct "methionine addiction" phenotype.This metabolic dependency arises from the aberrant demand and survival mechanisms of cancer cels for Met and its downstream metabolites. Consequently, methionine restriction (MR) has emerged as a promising therapeutic strategy in oncology. MR exerts its anti-tumor effects through multiple mechanisms: by disrupting S-adenosylmethionine (SAM)-dependent methylation reactions, impairing redox homeostasis, and suppressing polyamine synthesis.Notably,MR not only directly induces tumor cellcycle arrest and apoptosis but also remodels the tumor microenvironment and enhances anti-tumor immune responses. This review systematically summarizes the regulatory role of the Met metabolic pathway in tumor pathogenesis and the anti-tumor mechanisms of MR. It comprehensively reviews the latest research advances on the anti-tumor eficacy of MR in the treatment of various malignancies. Furthermore,it conducts an in-depth discussion on the clinical translation prospects and optimization strategies for MRbased therapies. The aim is to offer novel perspectives for developing metabolism-intervention-based precision cancer treatments.
