高通量环状RNA测序揭示hsacirc0001900在肾母细胞瘤中特异性高表达且与不良预后相关

Abstract: ObjectiveToexploretheexpressionprofileofcircularRNAs (circRNAs)andtheir potentialroles inprognosisand progression of Wilms'tumor (WT).MethodsFour pairs of WTandadjacent tissues were colectedfor high-throughput circRNAsequencingtoidentifythediferentiallyexpressedcircularRNAs.RT-qPCRwasusedtoverifytheexpressonlevels ofthetop6candidatecircRNAsintheclinicalsamples.hsa_circ0o900wasselectedforanalysisof itscorelationwith clinicopathologicalfeaturesandprognosisin34patients withWT.SangersequencingandRNaseRdigestionexperimentswere usedtoverifythecyclingsiteandstructural stabilityofhsa_circ_0o190molecule.ResultsAtotalof23978circularRNA moleculeswereidentifiedin WTtisuesbyhigh-throughputcircularRNA sequencing,andamong them 614were diferentiallexpressedinWT.hsa_circ0o9oshowedthehighestexpressionlevelamongthediferentiallyexpressed circRNAs,whichwasconsistentwiththefindingsinclinicaltumorsamplesandthesequencingresults.Corelationanalysis showedthathsacirco19oexpressionlevelwaspositivelycorrelatedwithWvolume,andthechildrenwithhighhsacic 0001900 expresion hadalowered recurrence-free survivalrate.The resultsof Sanger sequencing verifiedthe cicular splice sitesequenceof themolecule,andRnaseR digestionassayconfirmed itsstablecovalentstructure.Conclusion This study presentsacomprehensiveexpressnprofileofiularRNAsinWandtheexpreiolevelofhsa_cio9isatedto thesize ofWTandthepatients'prognosis,suggesting itspossibleroleasakeydriving gene inWTprogresion.
