Mechanism action turmeric on gout based on network pharmacology An Jingshuo.

【Abstract】Objective To predict the target turmeric's main active ingredients in the treatment gout, andtoexplore itsmulti-component,multi-target,multi-channel mechanism.MethodsThemain active components turmericwere screenedbythe TCM-PTD database,thepotential targets the above components were predicted bythe SwissTargetPrediction database,gout-related targets were obtained from the Human Genomes (GeneCards)databaseandthe CTD database.The target turmeric for gout was obtained,and theactive component-target network was constructed using Cytoscape 3.7.1 stware. The Gene Ontology (GO) and Kyoto Encyclopedia Genesand Genomes (KEGG) enrichment analysis were performed on the above targets using the Rlanguage package clusterPriler.Protein interaction network analysis target proteins was performed using BioGRID,InWeb_IM,andOmniPath databases.Visualizationand network topologyanalysiswereperformed by Cytoscapestware toobtain core targets.ResultsEight active ingredients,such as curcuminandbisabololwere obtained,which couldacton77targetsrelated togout.Theresults theGOenrichmentanalysisshowedthatthe above targets weresignificantly enriched inreactions involving metal ionsand oxidation.GO itemssuch asstress response:KEGG enrichment analysis showed that theabove targets weresignificantly enriched insignal pathways suchasFoxO signaling pathwayand interleukin(IL)-17 signaling pathway.ConclusionThe efect turmeric on goutreflects thecharacteristics multi-component,multi-targetandmulti-channel traditional Chinese medicine. It ishopedthatthisstudywillprovideascientificbasis forthescreeningactive ingredients inturmericand for further studies in pharmacology.
