【Abstract】ObjectiveTo explore themodeling efect low-dose intraperitoneal injection Lactobacilus casei cell wall extract （LCWE）emulsified with complete Freund'sadjuvant （CFA）to induce immunoglobulinA nephropathy （IgAN）model mice.MethodsThirty8-week-old C57BL/6J mice were romly divided intoa normal control group，a model group，a dapagliflozin group，with1O mice in each group.The Model group theDapa group were given multiple low-dose intraperitoneal injections LCWE emulsified with CFA.In the 1st 2nd weeks，the mixture LCWE CFA （0.5μg/g） was intraperitoneally injected 3 times a week， in the 3rd 4th weeks，the mixture LCWE CFA （0.5μg/g） was intraperitoneally injected1 time a week for a total 4 weeks.The Control group was given the same volume normal saline， the remaining operations remainedunchanged.Aftertheadministration，themice wereroutinelyraised for 4 weeks.Starting from the8th week，the Dapa group wasgiven intragastric administration dapagliflozin （Dapa）（10 mg/kg）.The modeling situation was observed after continuousadministration for 8 weeks.Urine specimens were colected to measure 24- hoururinaryprotein （24-UP）， blood was taken from theretro-orbital vein to detect thelevels serum creatinine （Scr） bloodurea nitrogen （BUN）.Renal tissue was taken for pathological staining to observe mesangial cells mesangial matrix， immunluorescence staining wasperformed to observethe mesangial area.ResultsCompared with the Control group，thelevels 24-urinary protein，serum Scr， BUN in the Model group increased.Compared with the Model group，the levels the above indicators in the Dapa group decreased， the differences were statistically significant （ P <0.05）. The results pathological histology staining immunluorescence staining showed that in the Model group，the mesangial matrix in therenal mesangial areaexped，mesangial cellsproliferated， IgA deposition increased.In the Dapa group，mesangial proliferation decreased，IgA deposition decreased.ConclusionLCWE can induce the expansion the glomerular mesangial matrix，the proliferation mesangial cels， the depositionIgAin themesangial area， can establish a stable IgAN animal model.
