Identification of characteristic genes in hepatocellular carcinoma epithelial cells and investigationoftheirmolecularmechanisms

[Abstract]Objective:Based on previous single-cellRNA sequencing (scRNA-seq) data revealing the high heterogeneityof hepatocelllarcarcinoma (HCC),thisstudyaimed to identifyandvalidate keydifferentially expressdgenes (DEGs)inmalignant epithelialcels,inordertoexploretheirpotentialasbiomarkersortherapeutictargets.Methods:Using previously established single-cell transcriptomicdataof HCC,weconducted in-depthanalyses focusing onmalignant epithelialcellpopulationsandscreenedforsignificantlydiferentiallyexpressedgenesthroughbioinformaticsapproaches. The expresion levelsof selected candidate genes were subsequently validated in hepatocellar carcinoma cellines using quantitativereal-time polymerasechain reaction (qRT-PCR).Results:The single-celldataanalysissuccessfully identified Neurotensin (NTS),fibrinogenbetachain(FGB),long-chainacyl-CoAsynthetase4(ACSL4),materallyexpresedene3 (MEG3),leptinreceptor(LEPR),and Janus kinase 3 (JAK3)askey genes with specifically high expression inmalignant epithelial celsoflivercancer.qRT-PCR validationresultswere highlyconsistent with thesequencingdata,confirming the aberrant expressionpatterns of these genes inHCCcellinesand pinpointingasubset of genes exhibiting stable dysregulationatboth transcriptionalandpost-transcriptional levels.Conclusion:Byintegrating single-cell transcriptomicanalysis with invitromolecularvalidation,thisstudyestablishedacompleteresearchloopfrom bioinformaticdiscoveryto experimentalconfirmation.Thevalidated DEGsprovideasolid molecular foundation forunderstanding thepathogenesisof HCC and ofer valuable evidence supporting their potential as diagnostic biomarkers or therapeutic targets.
