Expression of genes related to the cytoskeletal structure in gastric cancer cells and their role in the occurrence and development of gastric cancer

[Abstract]Objective:To investigate theexpresion characteristicsof molecular biomarkers ingastriccancercells andtheir roles in the occurrence and progresson of gastric cancer.Methods:The expression levels of genes such as PLEC,FAT1,EZR,HMGA1,MISPandPLAURinnormal humangastricepithelialcelsandgastriccancercellsweredetermined using real-time quantitative PCR(qRT-PCR) technology.The expresionsof PLECand PLAUR genes in MKN-1 and SGC-790lcelswererespectivelysilenced bygene interference technology,andtheproliferationabilitiesofcels in the si-PLEC,si-PLAUR and si-NC groups were determined by CCK8 method.Results:qRT-PCRresults showed that compared with normal human gastricepithelial cells,the expressonsofPLEC,FAT1,EZR,HMGA1,MISPandPLAUR genes in gastric cancer cells were significantly upregulated ( P <0.05).CCK8 experiment results showed that the cell reproductivecapacity in thesi-PLEC and si-PLAUR groups was significantly lower than that in the si-NC group ( P< (20 0.05).The tumorvolume formed by gastriccancercelswith PLEC expresion interference was significantlysmaler than that ofthecontrol group,andthetumor growthrate was significantlyslower.Conclusion:PLEC,FAT1,EZR,HMGA1, MISPand PLAURplayimportantroles intheoccurrenceanddevelopmentof gastriccancer,andmayserveaspotential molecular biomarkers for early diagnosis and targeted treatment.
