摘 要:目的:对当归的超临界CO2萃取-大孔树脂吸附联用技术的工艺进行研究。方法:当归的超临界CO2萃取,主要对萃取压力、萃取温度和萃取时间3个因素进行了正交考察,选用L9(34)正交表,以挥发油中主要成分藁本内酯为考察指标,优选出最佳萃取条件;对其萃取药渣采用水提-大孔树脂精制,以阿魏酸为指标,考察了树脂的种类、用量比例、醇洗脱的浓度及用量,优选出最佳的精制条件。结果:当萃取压力30MPa、温度为35℃,萃取2h,解析压力和温度分别为6Mpa和60℃时,当归的萃取物中挥发油得率为1.52%,其中藁本内酯的相对提取率为63.21%;而对萃取的药渣水提取后上AB8树脂,用量为以阿魏酸计为8.64mg/g干树脂,以4倍柱体积60%乙醇为洗脱溶媒时,阿魏酸的提取率(相对于水提液)为94.33%。结论:采用超临界CO2萃取-大孔树脂吸附工艺可以有效富集各成分部位,提高制剂载药量。

Fluid for Extracting- Macroporous Absorption Resin for Angelica sinensis (Oliv.)Diels

Abstract:Objective:To study the extraction technology on the application of supercritical CO2 fluid for extracting- macroporous absorption resin.Methods:Supercritical CO2 fluid extraction condition was carried out withL9(34) orthogonal designed with Z-ligustilide as standards. The extraction pressure,extraction temperature and extraction time were three factors in the experiment. The residue took the water extracts and macroporous absorption resin,employing the extracting ratio of ferulic acids as index,the effects of the resin types,the ratio,the concentration of ethanol and amount of ethanol on the extracting process were investigated. Results:The optimum extraction conditions were obtained:extraction pressure = 30Mpa,extraction temperature = 35℃,extraction time = 2 hours,separation pressure =6Mpa,separation temperature = 60℃.The AB-8 macroporous absorption resins to the residue extracts had the best separation efficiency when the volume of 60% ethanol as 4BV.Conclusion:This method established to optinize the extraction condition of volatile components of Angelica sinensis (Oliv.)Diels.
